Assay Principle
The kit measures Rubisco activity by tracking the oxidation of NADH to NAD+ at 340 nm. One molecule of ribulose-1,5-diphosphate (RuBP) combines with CO₂ to form two molecules of 3-phosphoglyceric acid (PGA), which is converted to glyceraldehyde-3-phosphate with concomitant NADH oxidation. The decrease in absorbance at 340 nm is directly proportional to Rubisco activity.
